目的 了解阿萨希毛孢子菌(Trichosporon asahii,T.asahii)菌丝生长与顶体(spitzenkper)的关系。方法 用荧光染剂FM4-64对YPD液体培养基培养后的T.asahii进行荧光染色,经不同浓度细胞松弛素D抑制后,在荧光显微镜下观察染色结果。结果 在T.asahii芽胞、芽管以及菌丝顶端和亚顶端均可见到FM4-64荧光聚集;随着细胞松弛素D浓度的增加,其顶体荧光逐渐消失。T.asahii的生长受到明显抑制。结论 T.asahii存在与真菌极性生长密切相关的顶体,其在控制菌丝生长方面起着重要的作用。
Objective To investigate the relationship between mycelium growth and spitzenkper in Trichosporon asahii (T.asahii). Methods Fluorescence staining of T.asahii cultured in YPD liquid medium with fluorescent dye FM4-64 was carried out. After being inhibited by different concentrations of cytochalasin D, the staining results were observed under a fluorescence microscope. Results FM4-64 fluorescence aggregates were observed in T.asahii sprouts, germ tubes and mycelium apical and apical apices. The acrosome fluorescence gradually disappeared with the increase of cytochalasin D concentration. T.asahii growth was significantly inhibited. Conclusion There is an acrosome closely related to polar growth of fungi in T.asahii, which plays an important role in controlling mycelial growth.
