为研究昆山接骨片对体外成骨细胞骨形成功能的影响 ,探讨昆山接骨片促进骨折愈合的机理 ,分别用 MTT法观察了含昆山接骨片 号和昆山接骨片 号的药物血清对成骨细胞增殖功能的影响 ,用 PNPP法观察昆山接骨片 号对成骨细胞碱性磷酸酶比活性的影响 ,以及用矿化结节计数法观察了昆山接骨片 号对成骨细胞矿化功能的影响。成骨细胞增殖率测定结果显示 ,昆山接骨片 号组为 0 .0 92± 0 .0 16 ,与对照组比较有显著性差异 (P<0 .0 5 ) ;而 号组为 0 .2 30± 0 .0 2 6 ,与对照组比较无差异。AL P比活性测定结果显示 ,昆山接骨片 号组为 0 .0 34± 0 .0 2 1,对照组为 0 .0 2 6± 0 .0 0 8,两组比较有显著性差异 (P<0 .0 5 ) )。矿化结节计数结果提示 ,昆山接骨片 号组为 2 5 .833± 7.782 ,对照组为 13.333± 6 .15 3,两组比较有显著性差异 (P<0 .0 5 )。综合研究结果表明 ,以活血化瘀药物为主制成的昆山接骨片 号具有促进成骨细胞的增殖、分化与矿化、成骨细胞的骨形成功能 ,从而发挥其促进骨折愈合的作用 ,单纯用螃蟹粉制成的昆山接骨片 号则无此作用
In order to study the effect of Kunshan Jiegu tablets on bone formation of osteoblasts in vitro, the mechanism of Kunshan Jiegu tablets in promoting fracture healing was discussed. MTT assay was used to observe the proliferation of osteoblasts in drug serum containing Kunshan Jiegu tablets and Kunshan Jiegu tablets. The effect of function was observed by PNPP method to observe the effect of Kunshan Jiegu tablets on the specific activity of alkaline phosphatase in osteoblasts, and the effect of Kunshan Jiegu tablets on mineralization of osteoblasts was observed by mineralized nodule counting method. The results of osteoblast proliferation rate showed that the Kunshan Jiegu Tablets group was 0.092±0.016, which was significantly different from the control group (P<0.05), while the number group was 0.23. ± 0 .0 2 6 , no difference from the control group. The results of the AL P specific activity test showed that the Kunshan Jiegu Tablets group was 0. 0 34 ± 0 . 0 2 1 and the control group was 0. 0 2 6 ± 0 . 0 0 8. There was a significant difference between the two groups (P< 0 .0 5))). The results of mineralized nodule counts showed that the Kunshan Jiegu Tablets group was 25.033±7.782 and the control group was 13.333±6.15. There was a significant difference between the two groups (P<0.05). The results of the comprehensive study indicate that the Kunshan Jiegu Tablets, which are mainly made by promoting blood circulation and removing stasis drugs, have the functions of promoting the proliferation, differentiation and mineralization of osteoblasts, and the osteogenic function of osteoblasts, thereby exerting its role in promoting fracture healing.
