用高效液相色谱法测定丙基硫氧嘧啶血药浓度。以咖啡因作内标,样品在酸性条件下采用氯仿—异丙醇混合溶剂进行抽提,于37℃水浴上吹干,残留物用甲醇溶解进行色谱分析。分析柱(250mm×4.6mm,ID)内填国产YWG-C_(18)固定相。流动相为含8%乙腈的O.025mol/L磷酸二氢钾水溶液。检测波长为272nm。用丙基硫氧嘧啶与内标的峰高比进行定量,其线性范围在0.1~4μg,(γ=0.9982),最低检测限可达0.05μg。本文还报道了6例初发甲亢病人体内丙基硫氧嘧啶药代动力学参数。
Determination of plasma concentration of propylthiouracil by high performance liquid chromatography. Using caffeine as internal standard, the sample was extracted under acidic conditions with chloroform-isopropanol mixed solvent, dried in a water bath at 37 ℃, and the residue was dissolved in methanol for chromatographic analysis. Analysis column (250mm × 4.6mm, ID) filled domestic YWG-C_ (18) stationary phase. The mobile phase consisted of O.025mol / L potassium dihydrogen phosphate aqueous solution containing 8% acetonitrile. The detection wavelength is 272nm. The peak area ratio of propylthiouracil to the internal standard was used to determine the linear range of 0.1 ~ 4μg (γ = 0.9982). The minimum detection limit was 0.05μg. This article also reported propylthiouracil pharmacokinetic parameters in 6 patients with initial hyperthyroidism.
