目的研究弹性蛋白酶诱导兔颈内动脉瘤模型的建立及其瘤壁平滑肌细胞表型变化。方法应用弹性蛋白酶作用于兔颈内动脉,建立新西兰兔颈内动脉瘤模型;分别于第1、2、3、4周取下实验动脉瘤标本行HE染色,逆转录PCR检测瘤壁平滑肌细胞表型基因(SM22α-mRNA)表达。结果实验动脉瘤随着不断生长瘤壁逐渐变薄,弹性纤维断裂,平滑肌细胞减少,SM22α-mRNA表达减少。结论弹性蛋白酶能稳定诱导动脉瘤的产生。动脉瘤动态生长过程中血管平滑肌细胞表型由收缩型向合成型转化。
Objective To investigate the establishment of rabbit model of intracranial aneurysm induced by elastase and the phenotypic changes of its smooth muscle cells. Methods The model of New Zealand rabbits’ internal carotid artery aneurysm was established by using elastase on the rabbit internal carotid artery. The specimens of experimental aneurysm were removed at 1, 2, 3 and 4 weeks respectively. HE staining and reverse transcription PCR were used to detect the expression of smooth muscle cell Type gene (SM22α-mRNA) expression. Results Experimental aneurysms with the growing wall gradually thinning, broken elastic fibers, smooth muscle cells decreased, SM22α-mRNA expression decreased. Conclusion Elastase can induce the aneurysm formation stably. Aneurysm during the dynamic growth of vascular smooth muscle cell phenotype from systolic to synthetic transformation.
