目的检测RAB5A、基质金属蛋白酶-9(MMP-9)在大肠癌组织中不同部位的表达及两者之间的相互关系。方法应用免疫组织化学SP法对46例大肠癌组织、距癌灶3cm的癌旁组织和23例非肿瘤患者大肠组织中RAB5A和MMP-9进行检测。结果46例大肠癌组织中,RAB5A蛋白的表达阳性率为100%(46/46),阳性表达位于胞质和胞膜;MMP-9的表达阳性率73.91%(34/46),阳性表达位于胞质。RAB5A和MMP-9表达在有淋巴结转移、Dukes分期晚及分化程度低的患者中表达程度高(P<0.05),MMP-9的表达还在大肠癌浸润度深的患者中表达程度高(P<0.05)。RAB5A和MMP-9的表达与性别、年龄、肿瘤部位均无关(P>0.05)。RAB5A和MMP-9的表达存在正相关(P<0.05,r=0.434)。结论RAB5A和MMP-9的表达有可能作为判断大肠癌恶性程度、转移的参考指标。
Objective To detect the expression of RAB5A and MMP-9 in different parts of colorectal carcinoma and the correlation between them. Methods Immunohistochemical SP method was used to detect RAB5A and MMP-9 in colorectal cancer tissues from 46 cases of colorectal cancer, 3cm away from the tumor and 23 cases of non-tumorous tissues. Results The positive expression rates of RAB5A protein in 46 cases of colorectal carcinoma were 100% (46/46). The positive expression of RAB5A protein was located in the cytoplasm and membrane of cells. The positive expression rate of MMP-9 was 73.91% (34/46) The expression is in the cytoplasm. The expression of RAB5A and MMP-9 was highly expressed in patients with lymph node metastasis, late Dukes staging and poorly differentiated (P <0.05), and the expression of MMP-9 was also high in patients with deep infiltration of colorectal carcinoma (P <0.05). The expression of RAB5A and MMP-9 had no correlation with sex, age and tumor location (P> 0.05). There was a positive correlation between the expression of RAB5A and MMP-9 (P <0.05, r = 0.434). Conclusion The expression of RAB5A and MMP-9 may be used as a reference index to judge the malignant degree and metastasis of colorectal cancer.
