目的建立测定不同产地连翘花中芦丁、连翘酯苷A、连翘苷、松酯酚含量的高效液相色谱法,为其有效成分控制提供有效方法。方法采用Symmetry C_(18)(4.6 mm×150 mm,5μm)色谱柱,流动相为乙腈-水,梯度洗脱,流速1.0 mL/min,检测波长230 nm,柱温25℃。结果不同产地的连翘花之间各成分的含量差异显著,其中连翘酯苷A在各样品中含量最高,芦丁含量次之,连翘苷含量最少;其中菏泽产的样品连翘酯苷A含量高达119.38±5.78 mg/g,连翘苷的含量达4.71±0.09 mg/g;临沂产的样品芦丁含量最高达75.64±2.09 mg/g;日照、青岛和烟台的样品中松脂酚含量较高且相差不大,分别是25.41±1.51、24.45±0.98、23.60±1.19 mg/g。结论 12个产地连翘花中连翘酯苷A的含量远远高于药典规定的最低含量要求。但连翘花中连翘苷的含量较低。
OBJECTIVE To establish a HPLC method for the determination of rutin, forsythoside A, forsythin, and pine ester in Forsythia suspensa (Forsythia) Flowers in different habitats, and to provide an effective method for the control of its active ingredients. Methods A Symmetry C_ (18) (4.6 mm × 150 mm, 5 μm) column was used. The mobile phase consisted of acetonitrile and water with a gradient of 1.0 mL / min. The detection wavelength was 230 nm and the column temperature was 25 ℃. Results Forsythia different from different regions of the composition of the content of significant differences, of which Forsythiaside A in each sample was the highest content of rutin followed by forsythin content of the least; which Heze sample forsythiaside The content of A was as high as 119.38 ± 5.78 mg / g and the content of forsythin as 4.71 ± 0.09 mg / g; the content of rutin in Linyi was up to 75.64 ± 2.09 mg / g; the contents of rosin in Rizhao, Qingdao and Yantai High and little difference, respectively, 25.41 ± 1.51,24.45 ± 0.98,23.60 ± 1.19 mg / g. Conclusions The content of Forsythiaside A in Forsythia suspensa from 12 habitats is much higher than the lowest requirement of pharmacopoeia. But forsythia flower forsythin content is low.
