为了开辟银杏种质保存新途径 ,以中南林学院经济林研究室选育的湖南梅核 号变异类型为材料 ,对银杏种胚超低温保存进行了研究 .结果表明 :银杏胚对脱水不太敏感 ,但胚含水量 (胚中水的质量与胚干物质质量的百分比 )低于 12 %时 ,试管苗生长发育异常 ;简单无菌风干燥脱水后的胚 ,液氮保存后恢复生长时 ,仅观察到子叶有限伸长、胚根生长、胚轴处产生大量白色的愈伤组织 ;无菌风干燥后的胚再经高浓度冰冻保护剂处理 ,超低温保存后再接种于培养基上 ,可产生再生植株
In order to open up a new way for the preservation of ginkgo germplasm and to study the cryopreservation of Ginkgo biloba germplasm using the variation type of Hunan Plum No. which was bred by the Forest Research Laboratory of Central South Forestry University, the results showed that: Ginkgo biloba was not sensitive to dehydration, When the embryo’s moisture content (percentage of embryo’s water quality and embryo’s dry matter mass) is less than 12%, the test tube seedlings grow abnormally; when the embryos are dehydrated in a simple aseptic air, Cotyledon limited elongation, radicle growth, hypocotyl generated a large number of white callus; sterile air dried embryos and then treated with high concentrations of cryoprotectants, cryopreserved and then inoculated on the medium, can produce regenerated plants
