本文首次报导了直立型扁蓿豆(Melilotoidesruthenicus(L.)Sojakcv.’Zhili’)成熟胚离体培养中器官发生的组织学研究。选用MS培养基并附加不同浓度的外源激素MA、NAA和BA进行培养,定期做冰冻切片进行观查。结果表明:IAA、NAA与BA的配合使用对直立型扁蓿豆成熟胚愈伤组织的诱导和不足芽的器官发生有显著的促进作用。不定芽起源于胚轴的皮层细胞,或表皮和皮层细胞同时启动。愈伤组织的形成可分为三个时期,启动期、分裂期、形成期;不定芽形成全过程可分为四个阶段:启动阶段、芽原基形成阶段、叶原基形成阶段和不定芽形态建成阶段。
This paper reports for the first time histological studies of organogenesis in mature embryos of upright-type alfalfa (Melilotoides ruthenicus (L.) Sojakcv.’Zhili ’). MS medium supplemented with different concentrations of exogenous hormones MA, NAA and BA were cultured regularly frozen sections were observed. The results showed that the combination of IAA, NAA and BA significantly promoted the induction of callus and the organ of under-buds in upright-type alfalfa beans. Adventitious buds originate from the cortical cells of the hypocotyl, or both epidermal and cortical cells start up. The formation of callus can be divided into three stages, the start-up period, the division stage and the formation stage. The whole process of adventitious bud formation can be divided into four stages: the start-up stage, bud primordium formation stage, leaf primordium formation stage and adventitious bud form stage.
