目的:利用近红外光谱法快速测定柴胡药材中柴胡皂苷a的含量。方法:利用HPLC测定柴胡样品中柴胡皂苷a的含量,运用偏最小二乘(PLS)法建立其含量与NIR光谱之间的多元校正模型,并对未知样品进行含量预测。结果:建立柴胡皂苷a定量模型准确性较好,内部交叉验证均方差(RMSECV)、内部交叉验证决定系数分别为0.321,0.934。经外部验证,NIR预测值与HPLC测定值之间的相关系数为0.931,预测回收率为103.84%。结论:利用近红外光谱法快速测定柴胡药材中柴胡皂苷a含量是可行的,该法可以为柴胡药材的快速无损检测提供参考和依据。
Objective: To rapidly determine the content of saikosaponin a in Radix Bupleuri by near-infrared spectroscopy. Methods: The contents of saikosaponin a in Bupleurum chinense were determined by HPLC. The multivariate calibration model was established by partial least squares (PLS) method and the content of saikosaponin was predicted. Results: The quantitative model of saikosaponin A was established with good accuracy, mean square error of internal cross-validation (RMSECV) and internal determination coefficients of cross validation of 0.321 and 0.944, respectively. Externally, the correlation coefficient between predicted NIR values and HPLC values was 0.931, with a predicted recovery of 103.84%. Conclusion: It is feasible to rapidly determine the content of saikosaponin a in Bupleurum by near-infrared spectroscopy. This method can provide reference and basis for rapid and nondestructive detection of Bupleurum.
