目的:探讨micro RNA-616(mi R-616)在肝细胞癌(HCC)中的表达及临床意义。方法:应用实时定量PCR检测80例HCC患者的肿瘤组织与癌旁组织标本,以及正常肝细胞与不同分化程度的HCC细胞mi R-616表达;分析mi R-616表达与HCC患者临床病理因素及预后的关系;观察HCC细胞过表达或抑制mi R-616表达后侵袭及转移能力的变化。结果:与癌旁组织比较,HCC组织中mi R-616的表达明显升高,且复发患者明显高于非复发患者,有转移患者明显高于无转移患者(均P<0.05);所有HCC细胞株的mi R-616表达均明显高于正常肝细胞,且在高侵袭性HCC细胞株中的表达明显高于低侵袭性HCC细胞株(均P<0.05);mi R-616表达与HCC患者是否存在门静脉癌栓、Edmondson-Steiner分级、TNM分期有关(均P<0.05);mi R-616表达是影响HCC患者生存的独立危险因素(P<0.05),mi R-616高表达患者的总生存率和无病生存率均明显低于mi R-616低表达患者(均P<0.05)。HCC细胞过表达mi R-616后侵袭迁移能力明显增强,mi R-616表达抑制后侵袭迁移能力明显减弱(均P<0.05)。结论:mi R-616在HCC中表达升高,且mi R-616高表达与HCC不良预后密切相关。
Objective: To investigate the expression of micro RNA-616 (mi R-616) in hepatocellular carcinoma (HCC) and its clinical significance. Methods: Real-time quantitative PCR was used to detect the expression of mi R-616 in 80 specimens of HCC from patients with HCC, normal liver cells and HCC cells with different degrees of differentiation. Prognosis; observed HCC cell overexpression or inhibition of mi R-616 expression changes in invasion and metastasis. Results: The expression of mi R-616 in HCC tissues was significantly higher than that in adjacent non-cancerous tissues, and the recurrence was significantly higher in patients with metastasis than in non-recurrence patients (all P <0.05). All HCC cells The expression of mi R-616 was significantly higher than that of normal hepatocytes, and the expression of mi R-616 in highly aggressive HCC was significantly higher than that in low invasive HCC (all P <0.05) (P <0.05). The expression of mi R-616 was an independent risk factor for survival in patients with HCC (P <0.05), and the overall expression of mi R-616 in patients with high expression of mi R-616 The survival rate and disease-free survival rate were significantly lower than those in patients with low expression of mi R-616 (all P <0.05). The invasion and migration ability of HCC cells was significantly increased after overexpression of mi R-616, and the ability of invasion and migration of mi R-616 was significantly weakened (all P <0.05). Conclusions: The expression of mi R-616 in HCC is increased, and the high expression of mi R-616 is closely related to the poor prognosis of HCC.
