使用国产仪器和试剂,一步法连续监测血清 LD 活力。NAD~+ 和 L-乳酸锂的 Km 分别为2.17×10~(-1)mmol/L 和 101.01×10~(-1)mmol/L,底物 NAD~+ 和 L-乳酸锂终浓度分别为4.30mmol/L 和 138.97mmol/L,反应速度为最大反应速度的89%,零级反应时间为6min,△A/min 在0.3以内具有良好的线性关系。同时,对连续监测的最适 pH 和标本的保存时间进行了实验探讨。100例健康献血员参考值为179±36.25u/L((?)±SD),与 Trace 试剂盒对比,两者显著相关(r=0.9792)。本法简便易行,具有临床实用价值。
Using domestic instruments and reagents, one-step continuous monitoring of serum LD activity. The Km of NAD~+ and L-Lactic acid lithium were 2.17×10 −1 mmol/L and 101.01×10 −1 mmol/L, respectively. The final concentrations of NAD~+ and L-lactate were respectively At 4.30 mmol/L and 138.97 mmol/L, the reaction rate was 89% of the maximum reaction rate, the zero-order reaction time was 6 min, and the △A/min was within a good linear relationship of 0.3. At the same time, experiments were conducted to investigate the optimum pH and specimen preservation time for continuous monitoring. The reference value of 100 healthy blood donors was 179±36.25u/L((?)±SD), which was significantly correlated with the Trace kit (r=0.9792). This method is simple and practical, with clinical practical value.
