目的:对超声波复合酶法中影响鲍氏针层孔菌菌丝体多糖提取效果的主要因素进行研究。方法:利用单因素试验、响应面分析和正交回归分析对提取工艺参数进行分析。结果:超声波提取胞内多糖优化工艺条件为:提取时间21 min,功率315W,料液比1∶25,在此条件下多糖得率为3.441%。在超声波优化结果基础上,进一步进行复合酶法处理,酶解最佳条件是:酶解时间120 min、pH5.5、酶解温度50℃、蛋白酶2%、纤维素酶3%、果胶酶1.5%,多糖得率为6.513%。结论:超声波和复合酶法双重处理提取桑黄胞内多糖是一种有效的提取方法,适合大规模生产运用。
OBJECTIVE: To study the main factors that affect the extraction efficiency of polysaccharide from P.citriclosporidium in ultrasonic complex enzyme. Methods: The extraction process parameters were analyzed by single factor test, response surface analysis and orthogonal regression analysis. Results: The optimal conditions of ultrasonic extraction of intracellular polysaccharides were: extraction time 21 min, power 315W, solid-liquid ratio 1:25, the yield of polysaccharide under this condition was 3.441%. Based on the results of ultrasonic wave optimization, the optimal enzymatic hydrolysis conditions were as follows: hydrolysis time 120 min, pH 5.5, enzymolysis temperature 50 ℃, protease 2%, cellulase 3%, pectinase 1.5%, polysaccharide yield was 6.513%. Conclusion: Ultrasound and enzyme complex treatment double extraction of Phellinus igniosa polysaccharide is an effective extraction method, suitable for large-scale production and use.
