目的探讨CD4~+CD25~+FOXP3~+调节性T细胞(CD4~+CD25~+FOXP3~+Treg)百分比的改变在哮喘小鼠发病中的作用。方法将雌性SPF级Balb/c小鼠20只随机分为哮喘模型组、正常对照组,哮喘模型组以鸡卵白蛋白(OVA)致敏,建立模型;正常对照组以生理盐水代替OVA。分别检测小鼠外周血(PB)与肺泡灌洗液(BALF)中嗜酸性粒细胞的数量及脾脏中CD4~+CD25~+FOXP3~+Treg的比例。结果哮喘组与对照组小鼠脾脏中CD4~+CD25~+/CD4~+(%)、CD4~+FOXP3~+/CD4~+(%)、CD4~+CD25~+FOXP3~+/CD4~+(%)分别为2.50±0.12与16.64±2.11、3.92±1.23与27.72±0.15、1.34±0.25与15.60±0.62,差异均有统计学意义(P<0.05)。哮喘小鼠脾脏CD4~+CD25~+FOXP3~+Treg占比较正常对照组明显减少。结论研究提示CD4~+CD25~+FOXP3~+Treg参与哮喘发病。
Objective To investigate the role of percentage change of CD4 ~ + CD25 ~ + FOXP3 ~ + regulatory T cells (CD4 ~ + CD25 ~ + FOXP3 ~ + Treg) in the pathogenesis of asthma in mice. Methods Twenty female SPF Balb / c mice were randomly divided into three groups: asthma model group, normal control group and asthmatic model group, which were sensitized with chicken ovalbumin (OVA). The normal control group was treated with normal saline instead of OVA. The number of eosinophils and the ratio of CD4 ~ + CD25 ~ + FOXP3 ~ + Treg in peripheral blood (PB) and BALF of mice were detected. Results The percentage of CD4 ~ + CD25 ~ + / CD4 ~ +, CD4 ~ + FOXP3 ~ + / CD4 ~ + and CD4 ~ + CD25 ~ + FOXP3 ~ + / CD4 ~ + (%) Were 2.50 ± 0.12 and 16.64 ± 2.11, 3.92 ± 1.23 and 27.72 ± 0.15, 1.34 ± 0.25 and 15.60 ± 0.62, respectively, with statistical significance (P <0.05). The percentage of CD4 ~ + CD25 ~ + FOXP3 ~ + Treg in spleen of asthmatic mice was significantly decreased as compared with the normal control group. Conclusions The study suggests that CD4 ~ + CD25 ~ + FOXP3 ~ + Treg is involved in the pathogenesis of asthma.
