目的 研究三氧化二砷 (As2 O3)对食管癌细胞增殖和热休克蛋白 70 (HSP70 )表达的影响。方法 通过相差显微镜、流式细胞术、免疫细胞化学染色和免疫印迹分析等方法观察As2 O3 对人食管癌细胞株EC1的作用效果和作用机制。结果 与对照组相比 ,经 2 μmol/L和 5 μmol/LAs2 O3 作用的细胞出现明显的生长抑制 ,G2 /M期细胞比例增加 ;2 μmol/LAs2 O3 作用 48h后EC1细胞HSP70 (heatshockprotein 70 )及HSC70 (heatshockcognateprotein 70 )表达均增加。结论 As2 O3 诱导食管癌细胞G2 /M期阻滞抑制细胞增殖和生长 ;HSP70的升高是细胞对As2 O3 作用后出现的应激反应 ,并与细胞周期阻滞相关。
Objective To study the effects of arsenic trioxide (As2 O3) on the proliferation of esophageal cancer cells and the expression of heat shock protein 70 (HSP70). Methods The effect and mechanism of As2 O3 on human esophageal carcinoma cell line EC1 were observed by phase contrast microscopy, flow cytometry, immunocytochemistry and Western blotting. Results Compared with the control group, the cells treated with 2 μmol / L and 5 μmol / L As 2 O 3 showed obvious growth inhibition and the proportion of cells in G2 / M phase increased. After treated with 2 μmol / L As 2 O 3 for 48 hours, heat shock protein 70 And HSC70 (heatshockcognateprotein 70) were increased. Conclusion As2 O3 induces G2 / M phase arrest in esophageal cancer cells, which inhibits cell proliferation and growth. The increase of HSP70 is a stress response after As2 O3 treatment and is associated with cell cycle arrest.
