目的研究脐血间充质干细胞(UCBMSC)对成人外周血淋巴细胞增殖的影响。方法分离、扩增UCBMSC并鉴定其表面标志及向成骨细胞及脂肪细胞的分化能力。UCBMSC与淋巴细胞直接共培养并用植物血凝素(PHA)刺激,或在TranswellTM非接触体系中,用PHA刺激共培养。两个培养体系均分为阴性对照组(淋巴细胞单独培养组)、阳性对照组(淋巴细胞加PHA刺激组)、实验组(淋巴细胞加PHA和UCBMSC组)。四氮唑衍生物MTS还原法测定细胞增殖并计算抑制率。结果在共培养与TranswellTM非接触体系中不同数量UCBMSC对淋巴细胞增殖均具有抑制作用,且UCBMSC浓度越高,抑制作用越强(P<0.05),具有剂量依赖性。但两体系中UCBMSC与淋巴细胞比值相同时,共培养体系抑制作用较非接触体系强,且差异具有统计学意义(P<0.05)。结论 UCBMSC能抑制淋巴细胞增殖,且共培养体系抑制作用较非接触体系强。
Objective To study the effects of umbilical cord blood mesenchymal stem cells (UCBMSCs) on the proliferation of adult peripheral blood lymphocytes. Methods UCBMSCs were isolated and expanded to identify their surface markers and their ability to differentiate into osteoblasts and adipocytes. UCBMSC is co-cultured directly with lymphocytes and stimulated with phytohaemagglutinin (PHA), or co-cultured with PHA in a Transwell (TM) non-contact system. The two culture systems were divided into negative control group (lymphocyte culture alone group), positive control group (lymphocyte plus PHA stimulation group), experimental group (lymphocyte plus PHA and UCBMSC group). Tetrazole derivatives MTS reduction assay of cell proliferation and calculate the inhibition rate. Results The proliferation of lymphocytes was inhibited by different numbers of UCBMSCs in the co-culture and TranswellTM non-contact systems. The higher the concentration of UCBMSC, the stronger the inhibitory effect was (P <0.05) in a dose-dependent manner. However, when the ratio of UCBMSC to lymphocyte was the same in both systems, the inhibitory effect of co-culture system was stronger than that of non-contact system, and the difference was statistically significant (P <0.05). Conclusion UCBMSC can inhibit the proliferation of lymphocytes, and the inhibitory effect of co-culture system is stronger than the non-contact system.
