目的 确定NF-kBp65在局灶性脑缺血再灌注后的空间及时间分布特征并测序。方法 采用免疫组化、RNA提取与RT-PCR技术及测序方法。结果 在脑缺血再灌注后不同时程及缺血边缘可见不同形态特征的神经元,在血脑屏障和血管周围的中性粒细胞也可见NF-kCBp65阳性反应。脑缺血2h再灌注后24h缺血侧NF-kBp65 mRNA RT-PCR的PCR产物相对量明显增高。结论NF-kBp65激活在局灶性脑缺血再灌注后的半暗区和血脑屏障中起重要作用。在恰当的时间窗内针对NF-kBp65的治疗可能是潜在的治疗途径。
Objective To determine the spatial and temporal distribution of NF-kBp65 after focal cerebral ischemia-reperfusion and to sequence it. Methods Immunohistochemistry, RNA extraction and RT-PCR techniques and sequencing methods were used. Results Neurons of different morphological characteristics were found at different time points after ischemia / reperfusion and at the edge of ischemia, and positive NF-κCBp65 was also seen in neutrophils around the blood-brain barrier and blood vessels. The relative amount of PCR product of NF-kBp65 mRNA RT-PCR on ischemia side 24h after reperfusion was significantly increased at 2h after cerebral ischemia. Conclusion Activation of NF-kBp65 plays an important role in the penumbra and blood-brain barrier after focal cerebral ischemia-reperfusion. Treatment for NF-kBp65 in the right time window may be a potential therapeutic approach.
