目的探讨慢性乙型肝炎病毒(HBV)感染过程中外周血单个核细胞(PBMC)的Toll样受体9(TLR9)表达水平与CD38、HLA-DR、CD95水平的关系。方法募集70例未经抗病毒治疗的慢性HBV感染者,按HBV DNA的含量高低分为高病毒载量组与低病毒载量组,12例健康人作为对照组;采集外周抗凝血,密度梯度法分离PBMC;使用流式细胞术检测PBMC的TLR9、CD38、HLA-DR、CD95的水平。结果慢性HBV感染过程中,高病毒载量组和低病毒载量组PBMC的TLR9、HLA-DR、CD95表达水平显著高于健康对照组;高病毒载量组和低病毒载量组PBMC上TLR9与CD38、HLA-DR、CD95的共表达率显著高于健康对照组;相关性分析结果显示慢性HBV感染者PBMC上,TLR9与CD38、HLA-DR、CD95的表达正相关(r分别为0.345、0.334、0.227)。结论慢性HBV感染过程中,PBMC的TLR9、HLA-DR、CD95表达水平升高,TLR9表达水平与CD38、HLA-DR、CD95水平正相关。
Objective To investigate the relationship between Toll-like receptor 9 (TLR9) expression and CD38, HLA-DR and CD95 levels in peripheral blood mononuclear cells (PBMCs) during the course of chronic hepatitis B virus infection. Methods Seventy patients with chronic HBV infection without antiviral therapy were divided into high viral load group and low viral load group according to the content of HBV DNA, and 12 healthy people as control group. The peripheral anticoagulant blood, density PBMCs were separated by gradient method. The levels of TLR9, CD38, HLA-DR and CD95 in PBMCs were detected by flow cytometry. Results The levels of TLR9, HLA-DR and CD95 in PBMC of high viral load group and low viral load group were significantly higher than those in healthy control group during chronic HBV infection. TLR9 The co-expression rate of CD38, HLA-DR and CD95 was significantly higher than that of healthy controls. Correlation analysis showed that the expression of TLR9 was positively correlated with the expression of CD38, HLA-DR and CD95 (r = 0.345, 0.334, 0.227). Conclusion The expression of TLR9, HLA-DR and CD95 in PBMC increased during chronic HBV infection, and the expression of TLR9 was positively correlated with the levels of CD38, HLA-DR and CD95.
