以玉米花粉为材料,提取总RNA后通过定量斑点杂交技术研究花粉发育过程中线 粒体nad4基因的转录规律.结果表明,在幼叶及不同发育时期的花粉中,线粒体nad4基 因的转录不存在其他线粒体基因那样的高度时空特异性;而是表现出等量转录规律.因此 我们认为花粉发育过程中线粒体基因的转录随基因种类、组织种类及植物种类而不同,或 表现时空特异性,或表现稳定的转录规律,同时对玉米不同发育阶段花粉总RNA的分离 技术进行了研究.
The total RNA was extracted from corn pollen, and the transcriptional regularity of mitochondrial nad4 gene during pollen development was studied by quantitative dot blot hybridization. The results showed that in young leaves and pollen at different stages of development, the mitochondrial nad4 gene transcription does not have the same high spatial-temporal specificity as other mitochondrial genes; Therefore, we think that the transcription of mitochondrial genes in pollen development varies with genotype, tissue type and plant species, or shows spatiotemporal specificity or stable transcriptome.
