目的:研究CD44粘附分子在颅内转移瘤及脑胶质母细胞瘤中的表达,探讨CD44粘附分子的表达与这些肿瘤发生转移及侵袭的关系。方法:用抗CD44粘附分子正常型(CD44s)蛋白单克隆抗体及抗CD44粘附分子变体型(CD44v-v10)蛋白多克隆抗体免疫组化染色检测10例正常脑组织、20例脑胶质母细胞瘤和20例颅内转移瘤中CD44粘附分子的表达。结果:10例正常脑组织中CD44s和CD44v3-v10表达均阴性(0/20);20例脑胶质母细胞瘤CD44s阳性表达率为100%(20/20),CD44v-v10表达阴性(0/20);20例颅内转移瘤CD44s表达阳性率为90%(18/20),CD44v-v10表达阳性率为70%(16/20)。CD44v-v10在颅内转移瘤及脑胶质母细胞瘤的表达有显著差异(P<0.01)。结论:①脑胶质母细胞瘤中无CD44变体蛋白表达可能,与其很少发生颅外转移有关;②CD44变体蛋白可能在颅内转移瘤向颅内转移的过程中起重要作用。CD44变体蛋白有可能成为颅内转移瘤诊治的有用指标之一。
OBJECTIVE: To study the expression of CD44 adhesion molecules in intracranial metastases and glioblastomas and to explore the relationship between the expression of CD44 adhesion molecules and the metastasis and invasion of these tumors. Methods: Immunohistochemical staining of polyclonal anti-CD44s monoclonal antibody and CD44v-v10 protein in 10 cases of normal brain tissue and 20 cases of glial Expression of CD44 Adhesion Molecules in B16 Tumors and 20 Cases of Intracranial Metastases. Results: The expression of CD44s and CD44v3-v10 in 10 normal brain tissues was negative (0/20). The positive rates of CD44s expression in 20 cases of glioblastoma were 100% (20/20) and 0 / 20). The positive expression rate of CD44s in 20 cases of intracranial metastases was 90% (18/20), and the positive rate of CD44v-v10 expression was 70% (16/20). The expression of CD44v-v10 in intracranial metastases and glioblastoma was significantly different (P <0.01). Conclusion: ① There is no expression of CD44 protein in glioblastoma, which may be related to the rare occurrence of extracranial metastasis. ②CD44 variant protein may play an important role in intracranial metastasis of intracranial metastases. CD44 variant protein may become a useful indicator of the diagnosis and treatment of intracranial metastases.
