花粉生活力是评估花粉细胞活性的依据之一,其活力测定结果的准确性决定细胞学试验和杂交育种的成败。本研究以32个百合品种的花粉为试验材料,采用染色法和花粉离体萌发法,探讨了不同百合品种新鲜花粉生活力、萌发率的差异,不同贮藏条件下同品种花粉生活力的差异。结果表明:离体萌发法的测定结果更能反映花粉活力,Casa Blanca的萌发率最高,为87.33%,Yelloween的萌发率最低,为1.33%。对于相同的贮藏条件即室温贮藏来说,因品种和贮藏时间的不同花粉生活力变化有明显差异,总的趋势为随贮藏时间延长,萌发率下降,但不同基因型间下降速度不同。在冷藏条件下,花粉萌发率随贮藏时间的推移总体也呈下降趋势,但下降速度比室温贮藏的明显缓慢,且不同品种的变化亦有所差别。
Pollen viability is one of the bases for assessing the activity of pollen cells. The accuracy of the determination of the viability of the pollen determines the success of the cytological test and the crossbreeding. In this study, the pollen of 32 lily varieties was used as the experimental material. The differences of fresh pollen viability and germination rate of different lily varieties and pollen viability under different storage conditions were investigated by the method of dyeing and in vitro germination of pollen. The results showed that the results of in vitro germination assay better reflected the pollen viability. Casa Blanca had the highest germination rate of 87.33%, while Yelloween had the lowest germination rate of 1.33%. For the same storage conditions that room temperature storage, due to different varieties and storage time pollen viability changes have significant differences, the general trend with the storage time, the germination rate decreased, but the rate of decline between different genotypes. Under cold storage conditions, the germination rate of pollen decreased with the storage time, but the decline rate was obviously slower than that stored at room temperature, and the changes of different varieties also differed.
