利用序列相关扩增多态性分子标记SRAP(Sequence-Related Amplified Polymorphism)技术对新疆梨40个栽培品种进行遗传多态性的研究。由http//genome.cs.mtu.edu/sas.html查得上游引物12条、下游引物14条,进行组合扩增筛选。对其中特异性强、稳定性好的10对引物(me2+em1、me3+em2、me5+em14、me6+em2、me7+em3、me7+em14、me10+em1、me11+em1、me11+em3、me11+em10)进行SRAP分析。10对引物共可扩增出131个条带,其中127条为多态性位点,平均每组引物扩增出13.1条多态性带。在物种水平上,平均多态性条带比率为96.95%。通过SRAP遗传关系聚类图聚类分析表明,在遗传相似系数0.625为标准40个梨种质分为3大类群:第一大类群中包括2份种质(砀山梨、奎克阿木特2号);第二大类群中包括6份种质(黄金、京白×砀山、白皮砀山、麻梨、新梨7号、句句梨);第三类群中包括其余32份种质。
The genetic polymorphism of 40 cultivars of pear in Xinjiang was studied by using Sequence-Related Amplified Polymorphism (SRAP). A total of 131 bands were amplified with 10 pairs of primers, of which 127 were polymorphic. A total of 13.1 polymorphic bands were amplified from each group of primers. At the species level, the average polymorphism banding ratio was 96.95%. Cluster analysis based on SRAP genetic relationship showed that the genetic similarity coefficient of 0.625 was divided into three groups according to the standard 40 pear germplasm: the first group consisted of 2 germplasm (Vulgaris pear, Quaker Amu 2 ); The second group includes 6 germplasm (gold, Jingbai × 砀 Hill, Pik Pik Hill, Pear, Xinli 7, sentence sentence pear); the third group includes the remaining 32 germplasm.
