目的探讨血吸虫虫卵抗原(SEA)对巨噬细胞RAW264.7和肺动脉内皮细胞(HPAECs)上清液中白细胞介素1β(IL-1β)表达的影响。方法体外培养RAW264.7细胞和HPAECs,随机分为对照组(A组)、脂多糖+SEA组(B组)和SEA组(C组),采用ELISA法检测细胞上清液中IL-1β表达量。结果与A组相比,B组和C组细胞上清液中IL-1β的表达均上调(P<0.05);C组IL-1β的表达低于B组(P<0.05)。结论 SEA诱导巨噬细胞和HPAECs上清液中IL-1β的表达,提示SEA诱导的炎症可能参与血吸虫病相关性肺动脉高压的发生。
Objective To investigate the effect of Schistosoma japonicum egg antigen (SEA) on the expression of interleukin-1β (IL-1β) in macrophage RAW264.7 and supernatant of pulmonary artery endothelial cells (HPAECs). Methods RAW264.7 cells and HPAECs were cultured in vitro and randomly divided into control group (A group), lipopolysaccharide + SEA group (B group) and SEA group (C group). The expression of IL-1β in cell supernatant the amount. Results Compared with group A, the expression of IL-1β in group B and group C was up-regulated (P <0.05). The expression of IL-1β in group C was lower than that in group B (P <0.05). Conclusions SEA induces IL-1β expression in macrophages and HPAECs supernatant, suggesting that SEA-induced inflammation may be involved in the pathogenesis of schistosomiasis-related pulmonary hypertension.
