目的:分析氯胺酮对SD幼鼠海马GAP-43蛋白表达的影响。方法:取7~10日龄的SD幼鼠36只,随机分为A、B、C三组各12只。A组注射生理盐水50 mg/kg,B组注射氯胺酮25 mg/kg,C组注射氯胺酮50 mg/kg。24 h后处死小鼠取海马组织,分别采用免疫组化与Western Blotting检测海马GAP-43蛋白的表达情况。结果:经图像分析软件处理后得知,A组、B组、C组条带灰度值分别为187.43±7.98、164.87±8.35、154.31±6.98,组间比较差异均有统计学意义(P<0.01)。A组、B组、C组GAP-43阳性细胞计数分别为(87.45±14.98)个/mm2、(66.48±12.36)个/mm2、(35.32±9.93)个/mm2,组间比较差异均有统计学意义(P<0.01)。结论:氯胺酮可明显降低SD幼鼠海马GAP-43蛋白的表达水平,这可能与其影响幼鼠神经系统发育有关。
Objective: To analyze the effect of ketamine on the expression of GAP-43 protein in hippocampus of SD rats. Methods: Totally 36 SD young rats aged 7-10 days were randomly divided into three groups (A, B and C). Group A received normal saline 50 mg / kg, group B received ketamine 25 mg / kg and group C received ketamine 50 mg / kg. Twenty-four hours later, the hippocampus was killed and the expression of GAP-43 protein in hippocampus was detected by immunohistochemistry and Western Blotting respectively. Results: After the image analysis software was processed, the gray values of the bands in group A, group B and group C were 187.43 ± 7.98, 164.87 ± 8.35 and 154.31 ± 6.98 respectively, with significant difference between the two groups (P < 0.01). The counts of GAP-43 positive cells in group A, group B and group C were (87.45 ± 14.98) /mm2, (66.48 ± 12.36) /mm2, (35.32 ± 9.93) / mm2, respectively Significance (P <0.01). Conclusion: Ketamine can significantly decrease the expression of GAP-43 protein in the hippocampus of SD rats, which may be related to its effect on the development of the nervous system in young rats.
