目的:探讨大肠癌中OCT4基因与CD44的关系。方法:收集2011年7月~2012年5月的32例大肠癌患者的癌组织和正常组织,采用qRT-PCR和Western blot的方法检测癌和正常肠上皮组织的OCT4和CD44的表达,并作相关性分析。结果:qRT-PCR检测癌组织中的OCT4的mRNA表达量为0.065±0.01,明显高于正常肠上皮组织的0.012±0.008(P<0.05);CD44s在癌组织中的表达也明显高于正常组织(0.023±0.008 vs0.009±0.002(P<0.05),经过spearman相关分析发现OCT4和CD44的mRNA表达存在正相关关系,r=0.587,P<0.05)。Western blot检测了其中2例患者的癌和正常组织的OCT4和CD44的蛋白表达,发现癌组织中两者的表达均较正常肠上皮表达高。结论:大肠癌中OCT4和CD44均呈高表达,两者呈正相关关系。
Objective: To investigate the relationship between OCT4 gene and CD44 in colorectal cancer. Methods: The cancer tissues and normal tissues from 32 patients with colorectal cancer from July 2011 to May 2012 were collected. The expressions of OCT4 and CD44 were detected by qRT-PCR and Western blot. Correlation analysis. Results: The mRNA expression of OCT4 in cancer tissue by qRT-PCR was 0.065 ± 0.01, which was significantly higher than that of normal intestinal epithelium (0.012 ± 0.008, P <0.05). The expression of CD44s in cancer tissues was also significantly higher than that in normal tissues (0.023 ± 0.008 vs 0.009 ± 0.002, P <0.05). There was a positive correlation between mRNA expression of OCT4 and CD44 after spearman correlation analysis (r = 0.587, P <0.05). The protein expressions of OCT4 and CD44 were detected by Western blot in two of the cancerous and normal tissues. The expression of both OCT4 and CD44 in cancerous and normal tissues was higher than that in normal intestinal epithelium. Conclusion: The expressions of OCT4 and CD44 in colorectal cancer are highly expressed, and the two are positively correlated.
