目的:比较不同采血方式对狂犬病疫苗免疫小鼠中和抗体检测的影响,为狂犬病研究中选择更适合抗狂犬病病毒中和抗体(rabies virus neutralizing antibody,RVNA)检测的采血方法提供科学依据。方法:将50只4~6周龄雌性昆明小鼠随机分为3组,A组(摘眼球采血,间隔7天,n=30)、B组(内眦采血,间隔7 d,n=10)、C组(内眦采血,间隔14 d,n=10),各组小鼠进行狂犬病疫苗免疫后,于不同时点取血检测血清抗体,分析不同采血方式对小鼠RVNA的影响。每日观察小鼠状态,并记录体重。结果:在整个试验中,各组小鼠采血前后体重均有正常性增长,但各组体重差异无统计学意义(n P>0.05);小鼠利用率A组高于B、C组,B组和C组小鼠利用率差异无统计学意义(n P>0.05);各组小鼠RVNA水平之间差异均无统计学意义(n P>0.05)。n 结论:在小鼠体重、RVNA水平无差异的前提下,综合考虑试验成本、动物福利等因素的影响,认为间隔7天内眦采血法为检测RVNA水平试验最合适的采血方法。“,”Objective:To compare the effects of different blood collection methods on the detection of neutralizing antibodies in rabies vaccine immunized mice, so as to provide scientific basis for selecting blood collection methods more suitable for the detection of anti-rabies virus neutralizing antibody (RVNA) in rabies research.Methods:Fifty female Kunming mice aged 4-6 weeks were randomly divided into 3 groups, including group A (eyeball blood collection, 7 days interval, n=30), group B (inner canth blood collection, 7 days interval, n=10) and Group C (intracanthus blood sampling, 14 days interval, n=10), After immunization with rabies vaccine in each group of mice, blood was taken at different times to detect serum antibodies, and the effects of different blood collection methods on the detection of neutralizing antibodies in mice were analyzed. The status and the body weight of the mice were recorded daily.Results:Throughout the experiment, the weight of mice before and after blood collection in each group had a normal increment every day, but the changes were not statistically significant (n P>0.05). The utilization rate of mice in group A was higher than that in groups B and C. There was no statistical difference in the utilization rate of mice in groups B and C (n P>0.05). There was no statistical difference between the RVNA levels of mice in each group (n P>0.05).n Conclusions:On the premise of no differences in weight of mice and RVNA levels, and with comprehensive consideration on the impact of test cost, animal welfare and other factors, the candid blood collection method with 7 days interval is recommended as the most suitable blood collection method for detecting RVNA level.
