目的研究TGFα,EGFR反义寡聚核苷酸抑制大肠癌HR8348细胞株恶性增殖的作用.方法采用23个碱基组成的TGFα反义寡聚核苷酸和21个碱基组成的EGFR反义寡聚核苷酸以及21个碱基组成的对照寡聚核苷酸作用于人大肠癌HR8348细胞,通过细胞生长抑制实验,3HTdR掺入,mRNA斑点杂交及细胞周期分析以确定其对大肠癌细胞的增殖抑制作用和作用环节.结果TGFα,EGFR反义寡聚核苷酸与对照寡聚核苷酸相比较,均能有效抑制HR8348细胞的增殖(P<001),DNA合成(P<001)和mRNA(P<001)的表达,并能有效地延缓HR8348细胞由G0/G1期向S期的转化,延缓了细胞的分裂增殖
Objective To study the effect of TGFα and EGFR antisense oligonucleotides on malignant proliferation of colorectal cancer HR8348 cell line. Methods Human colon cancer HR8348 was treated with 23 base TGFα antisense oligonucleotides, 21 bases EGFR antisense oligonucleotides and 21 bases control oligonucleotides. Cells, through cell growth inhibition experiments, 3H TdR incorporation, mRNA dot blot hybridization and cell cycle analysis to determine its inhibition of proliferation of colorectal cancer cells and its role in the process. Results Compared with control oligonucleotides, TGFα and EGFR antisense oligonucleotides could effectively inhibit the proliferation of HR8348 cells (P<001), DNA synthesis (P<001) and mRNA (P). The expression of <001) can effectively delay the transformation of HR8348 cells from G0/G1 phase to S phase, delaying the proliferation of cells.
