目的检测口咽癌患者的HPV-16和HPV-18感染情况,了解不同HPV型别在口咽癌中的分布特点及相关性分析。方法本研究设置实验组和对照组,实验组为71例口咽癌标本,对照组为83例非口咽癌标本。在本院病理科切取蜡块标本,脱蜡,用加碱煮沸法提取DNA后进行PCR扩增,最后进行导流斑点杂交法杂交,实验结果采用SPSS22.0分析软件统计。结果 71例口腔癌标本中有16例检出HPV,其中检出8例HPV-16(占所有型别的50%),5例HPV-18(占所有型别的31.25%),2例HPV-11和1例HPV-33;83例正常标本内均未检出HPV感染。实验组中HPV-16和HPV-18的感染率差异无统计学意义(P=0.390)。结论口咽癌患者中HPV-16和HPV-18的检出率之间没有显著性差异,这与有关文献报道不同,可能为地域差异引起的。
Objective To detect the infection of HPV-16 and HPV-18 in patients with oropharyngeal carcinoma and investigate the distribution and correlation of different HPV types in oropharyngeal carcinoma. Methods The experimental group and control group were set up in this study. The experimental group consisted of 71 cases of oropharyngeal carcinoma and the control group of 83 cases of non-oropharyngeal carcinoma. Paraffin wax specimens were taken from the pathology department of our hospital and dewaxed. The DNA was extracted with alkali and boiled and then amplified by PCR. Finally, the hybridization was performed by dot blot hybridization. The experimental results were analyzed by SPSS 22.0 software. Results HPV was detected in 16 out of 71 oral carcinomas, including 8 cases of HPV-16 (50% of all cases), 5 cases of HPV-18 (31.25% of all cases), 2 cases of HPV -11 and 1 case of HPV-33; HPV infection was not detected in 83 cases of normal specimens. The infection rate of HPV-16 and HPV-18 in experimental group had no significant difference (P = 0.390). Conclusion There is no significant difference between the detection rate of HPV-16 and HPV-18 in patients with oropharyngeal carcinoma, which is different from the reported in the literature and may be caused by geographical differences.
