目的:在体外研究高糖刺激人肾脏间质成纤维细胞时,HGF、TGF-β的表达以及黄芪延缓糖尿病肾病与其的相关性。方法:将人肾脏间质成纤维细胞分别培养于不同浓度的葡萄糖中,于 24 h后用 RT-PCR方法检测HGF mRNA的转录水平,继而用不同浓度的 HGF刺激细胞来现察 TGF-βmRNA的表达。随即用黄芪药物血清刺激细胞,观察HGF的表达情况。结果:高浓度葡萄糖培养的细胞早期就有HGF表达,之后表达逐渐下降,而TGF-β在此之后出现表达上升。同时外源性HGF可以抑制TGF-βmRNA的表达。黄芪含药血清可以显著刺激细胞表达HGF。结论:高糖刺激后可以诱导HGF表达,该表达的上升是一种有益的防御反应。此外,外源性加入HGF可以降低TGF-β的表达,从而促进细胞外基质的降解。黄芪通过诱导HGF的产生,起到抗纤维化的作用,从而延缓糖尿病肾病的进展。
OBJECTIVE: To investigate the expression of HGF, TGF-β and the relationship between Astragalus and diabetic nephropathy in high glucose-induced human renal interstitial fibroblasts. METHODS: Human renal interstitial fibroblasts were cultured in different concentrations of glucose, and the mRNA level of HGF mRNA was detected by RT-PCR after 24 h. Then, cells were stimulated with different concentrations of HGF to observe the expression of TGF-β mRNA. expression. The cells were immediately stimulated with Astragalus drug serum and the expression of HGF was observed. RESULTS: HGF was expressed early in cells incubated with high concentrations of glucose, and then gradually decreased, whereas TGF-beta expression increased thereafter. At the same time, exogenous HGF can inhibit the expression of TGF-β mRNA. Astragalus medicated serum can significantly stimulate the cells to express HGF. CONCLUSION: HGF expression can be induced after high glucose stimulation, and this rise in expression is a beneficial defense response. In addition, exogenously added HGF can reduce the expression of TGF-β, thereby promoting the degradation of extracellular matrix. Astragalus plays an anti-fibrotic effect by inducing the production of HGF, thereby delaying the progression of diabetic nephropathy.
