目的探讨脂多糖(LPS)激活小胶质细胞(MG)对黑质损伤多巴胺(DA)能神经元细胞存活的影响。方法采用立体定向技术向大鼠单侧黑质(SNpc)内注入LPS激活MG后,隔48h于原位注射高、低剂量鱼藤酮(RT)分别建立重度及轻度损伤模型;采用免疫组化法观察黑质酪氨酸羟化酶(TH)阳性细胞和离子钙结合转接分子1(Ibα-1)阳性MG变化。结果 LPS处理后低剂量RT组(A2)酪羟化酶(TH)阳性细胞数较只给同剂量RT(B2)组多(P<0.05),形态较(B2)组完整;予LPS处理后给高剂量鱼藤酮组(A3)较只给同剂量RT(B3)组DA神经元死亡增多(P<0.05),形态较(B3)组损伤重。结论 LPS激活MG对轻度损伤DA能神经元具有保护作用。
Objective To investigate the effect of lipopolysaccharide (LPS) activated microglia (MG) on the survival of dopaminergic neurons in substantia nigra of substantia nigra. Methods Stereotaxic technique was used to establish the model of severe and mild injury by injecting LPS into rat unilateral substantia nigra (SNpc) and then injecting high dose and low dose of rotenone (RT) into rats at 48h intervals. Immunohistochemistry The changes of nigral-tyrosine hydroxylase (TH) -positive cells and ion-binding calcium ion exchanger (Ibα-1) -positive MG were observed. Results After LPS treatment, the positive cells of tyrosinase (TH) in low dose RT group (A2) were more than those in the same dose of RT (B2) group (P <0.05) Compared with group B3, the death of DA neurons in high dosage rotenone group (A3) increased (P <0.05) compared with the same dose of RT (B3) group only. Conclusion LPS-activated MG can protect DA neurons with mild damage.
