目的研究少突胶质细胞肿瘤染色体1p/19q杂合性缺失(LOH)情况。方法采用即时荧光定量PCR微卫星分析技术对28例少突胶质细胞肿瘤进行染色体1p/19qLOH检测。结果28例少突胶质细胞肿瘤染色体中有24例(85·7%)发生1pLOH;有18例(64·3%)发生19qLOH;有17例(60·7%)发生1p/19q联合性LOH;25例有1p或19qLOH。结论即时荧光定量PCR微卫星分析技术特异性高、方便快捷,可以用于石蜡包埋组织标本染色体杂合性缺失的检测。大多数少突胶质细胞肿瘤发生了1p/19qLOH。
Objective To study the heterozygosity deletion (LOH) of chromosome 1p / 19q in oligodendrocyte. Methods Chromosome 1p / 19qLOH was detected in 28 oligodendrocyte neoplasms using real-time fluorescence quantitative PCR microsatellite analysis. Results Twenty-four cases (85.7%) of oligodendrocyte tumors had 1pLOH, 18 cases (64.3%) had 19qLOH, and 17 cases (60.7%) had 1p / 19q union LOH; 25 cases have 1p or 19qLOH. Conclusion Real-time PCR-based microsatellite analysis is highly specific, convenient and rapid and can be used to detect chromosome heterozygosity in paraffin-embedded tissue samples. Most oligodendrocyte tumors develop 1p / 19qLOH.
