目的 通过观察生长激素释放激素 (GHRH)、山德定、生长激素 (GH)和胰岛素样生长因子Ⅰ (IGF Ⅰ )对IM 9细胞中GH基因启动子活性的影响 ,以了解这些激素对B淋巴细胞中GH表达的调节作用。方法 把人外周血淋巴细胞GH基因上游 4 84~ + 2bp调控序列克隆到荧光素酶报告基因质粒中 ,然后把克隆得到的质粒转染到IM 9细胞内 ,通过测量荧光素酶的活性观察垂体GH的各种调节激素对GH基因启动子活性的影响。结果 0 .0 0 1nmol/L的人GHRH能够显著抑制荧光素酶在IM 9细胞中的表达 (P <0 .0 0 2 ) ,但 0 .1nmol/L和 1 0nmol/L的GHRH对荧光素酶的抑制作用反而减弱。人IGF Ⅰ ( 1、1 0和 1 0 0nmol/L)可增强荧光素酶的表达 (均P <0 .0 5)。GH( 1 .0ng/ml)可显著增加荧光素酶的表达 ,但作用的强度不大。不同浓度的生长抑素类似物山德定对IM 9细胞中荧光素酶的表达均无显著影响。结论 GHRH、IGF Ⅰ和GH对IM 9细胞中GH启动子的活性有一定的调节作用 ,说明调节垂体GH分泌的GHRH、IGF Ⅰ和GH等能够调节B淋巴细胞内irGH的表达。
OBJECTIVE: To investigate the effects of GHRH, SD, GH and IGF Ⅰ on GH promoter activity in IM 9 cells in order to understand the effect of these hormones on B lymph Modulation of GH expression in cells. Methods The regulatory sequences of 4 84 ~ +2 bp upstream of GH gene in human peripheral blood lymphocytes were cloned into luciferase reporter plasmids, and then the cloned plasmids were transfected into IM 9 cells, and the activity of luciferase was measured to observe the expression of pituitary Effects of GH Regulators on GH Gene Promoter Activity. RESULTS: Human GHRH at 0.010 nmol / L significantly inhibited the expression of luciferase in IM 9 cells (P <0. 02), but the effect of 0.1 nmol / L and 10 nmol / L of GHRH on the expression of fluorescein Enzyme inhibition instead weakened. Human IGF Ⅰ (1,1 0 and 100 nmol / L) enhanced luciferase expression (all P <0.05). GH (1 .0ng / ml) significantly increased luciferase expression, but the intensity of the effect was not significant. Sandostatin, a somatostatin analogue of different concentrations, had no significant effect on luciferase expression in IM 9 cells. Conclusion GHRH, IGF Ⅰ and GH can regulate the GH promoter activity in IM 9 cells, indicating that GHRH, IGF Ⅰ and GH, which regulate the secretion of pituitary GH, regulate the expression of irGH in B lymphocytes.
