目的探讨中药复方育泡饮对实验性自身免疫性卵巢早衰(POF)小鼠卵巢功能的影响。方法以成熟的SD雌性大鼠卵巢组织制备卵巢抗原,皮下多点注射免疫BALB/C雌性小鼠,建立卵巢早衰模型。在3次免疫后连续灌服中药复方,以强的松为阳性对照,酶联免疫法检测外周血雌二醇水平(E2)、卵泡刺激素(FSH)、外周血抗卵巢抗体(AoAb),IL-4、IL-10,观察卵巢的形态学变化,并计数各级卵泡及黄体数目。结果POF小鼠外周血E2下降,FSH升高,AoAb阳性,IL-4、IL-10明显提高,卵巢内有大量淋巴细胞浸润,各级卵泡及黄体数目减少。育泡饮能提高体内E2水平,减缓POF小鼠外周血FSH升高,抑制AoAb产生,降低IL-4与IL-10水平,减轻卵巢内淋巴细胞浸润,使各级卵泡及黄体数增加,育泡饮高剂量组比中剂量、低剂量组作用显著。结论育泡饮能减轻卵巢自身免疫性炎症损伤,对自身免疫性卵巢早衰具有治疗作用。
Objective To investigate the effect of Yufuyin, a traditional Chinese compound compound, on ovarian function in mice with experimental autoimmune premature ovarian failure (POF). Methods Ovarian antigen was prepared from the ovarian tissue of mature SD female rats. BALB/C female mice were immunized with multiple subcutaneous injections to establish a model of premature ovarian failure. After 3 immunizations, rats were continuously fed with Chinese herbal compound and prednisone was used as a positive control. Enzyme-linked immunosorbent assay was used to measure peripheral blood estradiol level (E2), follicle stimulating hormone (FSH), and peripheral blood anti-ovarian antibody (AoAb). The morphological changes of the ovary were observed with IL-4 and IL-10, and the number of follicles and corpus luteum were counted. Results The peripheral blood E2 decreased, FSH increased, AoAb was positive, IL-4, IL-10 were significantly increased in POF mice. There was a large number of lymphocyte infiltration in the ovary, and the number of follicles and corpus luteum decreased at all levels. Yu Pao Yin can increase the level of E2 in the body, slow the increase of FSH in peripheral blood of POF mice, inhibit the production of AoAb, reduce the levels of IL-4 and IL-10, reduce the infiltration of lymphocytes in the ovary, and increase the number of follicles and corpus luteum at all levels. High-dose Beverage Group had a significant effect on the middle-dose and low-dose groups. Conclusion Yufu decoction can reduce autoimmune inflammatory injury in the ovary and has therapeutic effect on autoimmune premature ovarian failure.
