目的:观察精子蛋白17(Sp17)的异常表达对体外培养的卵巢癌细胞顺铂和卡铂耐药性的影响。方法:将Sp17基因转染卵巢癌细胞系HO8910,采用流式细胞术测定目的蛋白的表达,获得稳定高表达Sp17的卵巢癌细胞系HO8910/Sp17。MTT法检测铂类药物对转染细胞HO8910/Sp17的生长抑制作用并与未转染的亲代细胞进行比较。结果:在有效剂量范围内,顺铂和卡铂对HO8910/Sp17细胞的抑制率明显低于亲本细胞HO8910,P<0.05。耐药指数顺铂1.4,卡铂1.38。结论:Sp17明显增加卵巢癌细胞对铂类化疗药物的耐药性。
Objective: To observe the effect of abnormal expression of sperm protein 17 (S17) on cisplatin and carboplatin resistance in cultured ovarian cancer cells. Methods: The ovarian cancer cell line HO8910 was transfected with Sp17 gene. The expression of the target protein was determined by flow cytometry. The ovarian cancer cell line HO8910 / Sp17 stably expressing Sp17 was obtained. The growth inhibition of the transfected cells HO8910 / Sp17 was detected by MTT assay and compared with untransfected parental cells. Results: The inhibitory rates of cisplatin and carboplatin on HO8910 / Sp17 cells were significantly lower than those of HO8910 cells in the effective dose range (P <0.05). Drug resistance index cisplatin 1.4, carboplatin 1.38. Conclusion: Sp17 significantly increases the resistance of ovarian cancer cells to platinum chemotherapeutic drugs.
