目的:研究短程大剂量粒细胞集落刺激因子(G-CSF)对外周血造血干细胞的动员作用。方法:采用短程大剂量G-CSF对11例患者进行外周血造血干细胞动员,G-CSF5μg/kg皮下注射,每日2次,共3天,动员当天及第4天(单采日),分别取骨髓及外周血进行常规检查,CFU-GM、BFU-E、CFU-mix集落培养及CD34、CD38细胞检测。结果:动员后患者骨髓增生明显活跃,外周血白细胞计数明显升高。骨髓、外周血CFU-GM、BFU-E及CFU-mix集落,CD34+CD38+细胞、CD34+CD38-细胞均较动员之前明显增多(P<0.05)。一次单采的单个核细胞(MNC)平均为1.56×108/kg。冷冻后MNC、CFU-GM、BFU-E及CD34+CD38+细胞的恢复率为80%(P>0.05);CFU-mix的恢复率为60%,但CD34+CD38-细胞冷冻后减少(P<0.05)。回输患者体内MNC的平均集落数分别为:CFU-GM6.70×104/kg;BFU-E1.60×104/kg;CFU-mix0.32×104/kg;CD34+CD38+细胞1.60×106/kg;CD34+CD38+细胞0.23×106?
AIM: To investigate the mobilization of peripheral blood hematopoietic stem cells by short-term high-dose granulocyte-colony stimulating factor (G-CSF). Methods: 11 cases of peripheral blood hematopoietic stem cell mobilization and 5-g / kg G-CSF subcutaneously were injected subcutaneously twice a day for 3 days with short-range and high-dose G-CSF. The mobilization day and the fourth day Bone marrow and peripheral blood were taken for routine examination, CFU-GM, BFU-E, CFU-mix colonies and CD34, CD38 cells. Results: Myeloid hyperplasia was significantly active after mobilization, and the peripheral blood leukocyte count was significantly increased. CFU-GM, BFU-E and CFU-mix colonies, CD34 + CD38 + cells and CD34 + CD38- cells in bone marrow and peripheral blood were significantly increased compared with before mobilization (P <0.05). A single apheresis mononuclear cells (MNC) averaging 1.56 × 108 / kg. The recovery rate of MNC, CFU-GM, BFU-E and CD34 + CD38 + cells after cryopreservation was 80% (P> 0.05). The recovery rate of CFU-mix was 60%, but the number of CD34 + CD38- cells decreased after freezing (P < 05).
