目的观察钬激光照射后对人膀胱癌细胞株T24增殖活性的影响与诱导细胞凋亡的作用。方法取对数生长期人膀胱癌细胞株T-24细胞,分对照与实验两组,实验组以800mJ钬激光直接照射而对照组不激发能量直接照射,于照射后48h分别选用光镜下直接观察细胞形态学的改变,流式细胞仪检测及免疫细胞化学检测等方法研究其对细胞增殖及凋亡的影响。结果800 mJ钬激光照射后实验组细胞增殖明显受抑制,细胞形态学及流式细胞学检测均呈现出凋亡改变,免疫细胞化学检测对照组和实验组半胱氨酸蛋白酶-3(Caspase-3)蛋白阳性细胞平均积分吸光度值分别为114.23±2.69,159.40±1.98,其差异有统计学意义(P<0.05)。结论体外细胞试验结果表明,采用钬激光照射人膀胱癌细胞株T-24,能够抑制肿瘤细胞增殖活性,诱发肿瘤细胞凋亡可能是钬激光抑制作用的机制之一。
Objective To observe the effects of holmium laser irradiation on the proliferation of human bladder cancer cell line T24 and the induction of apoptosis. Methods The human bladder cancer cell line T-24 cells in logarithmic growth phase were divided into control and experimental groups. The experimental group received direct irradiation with 800mJ holmium laser while the control group did not stimulate the energy directly. After 48h irradiation, To observe the changes of cell morphology, flow cytometry and immunocytochemistry detection of cell proliferation and apoptosis. Results After 800 mJ holmium laser irradiation, the proliferation of the experimental group was significantly inhibited. The morphological changes of the cells were observed by flow cytometry. The expressions of caspase- 3) protein positive cells were 114.23 ± 2.69,159.40 ± 1.98, the difference was statistically significant (P <0.05). Conclusion The results of in vitro experiments show that holmium laser irradiation of human bladder cancer cell line T-24 can inhibit tumor cell proliferation and induce apoptosis of tumor cells, which may be one of the mechanisms of holmium laser inhibition.
