目的探讨复方中药对老年大鼠心肌细胞间隙连接蛋白43(connexin 43,Cx43)表达的影响,为心脏疾病的中药治疗提供细胞生物学及分子生物学基础。方法利用免疫组织化学和图像分析方法探讨复方中药处理对大鼠心肌细胞Cx43的分布和表达量的影响。结果免疫组化结果显示,空白对照组Cx43呈点状,散布在心肌细胞胞浆中;复方中药组Cx43呈线状,多分布在心肌细胞闰盘处和侧-侧相接处。图像分析显示复方中药组和空白对照组对Cx43的表达量影响的差异无统计学意义(P>0.05)。结论复方中药处理后,可改善老年大鼠心肌细胞Cx43分布。
Objective To investigate the effects of compound Chinese herbs on the expression of connexin 43 (Cx43) in the aged rats and provide the basis of cell biology and molecular biology for the treatment of heart diseases with traditional Chinese medicine. Methods Immunohistochemistry and image analysis were used to investigate the effects of compound Chinese herbs on the distribution and expression of Cx43 in rat cardiomyocytes. Results The results of immunohistochemistry showed that Cx43 was distributed in the cytoplasm of cardiomyocytes in the blank control group. Cx43 was linear in the compound Chinese herbal medicine group and distributed in the intercalary cardiocytes and the lateral-lateral junction. Image analysis showed that there was no significant difference in the expression of Cx43 between the compound traditional Chinese medicine group and the blank control group (P> 0.05). Conclusion The compound Chinese medicine treatment can improve the distribution of Cx43 in rat myocardial cells.
