目的探讨IL-17、IFN-γ在中耳胆脂瘤中的表达水平及其在骨质破坏中的意义。方法用免疫组织化学、ELISA法及RT-PCR的方法检测27例中耳胆脂瘤、23例中耳炎肉芽和19例正常外耳道皮肤组织中的IL-17、IFN-γ蛋白和mRNA的表达水平,并分析其表达与胆脂瘤骨质破坏的相关性。结果 IL-17在中耳胆脂瘤上皮下基质内呈强阳性表达,明显高于肉芽组织和外耳道皮肤,差异有统计学意义(P<0.05);IFN-γ在中耳胆脂瘤上皮下基质内呈强阳性表达,明显高于外耳道皮肤,与肉芽组织比较,含量差异无统计学意义(P>0.05)。胆脂瘤中IL-17 mRNA的表达量高于肉芽组织、外耳道皮肤;胆脂瘤中IFN-γmRNA的表达量高于与肉芽组织、外耳道皮肤,差异有统计学意义(P<0.05);中耳胆脂瘤中IL-1、IFN-γ的表达量与骨质破坏程度呈正相关(P<0.01)。结论中耳胆脂瘤组织中的IL-17、IFN-γ表达水平上调,且与该病骨质破坏的程度有关。
Objective To investigate the expression of IL-17 and IFN-γ in middle ear cholesteatoma and its significance in bone destruction. Methods The expression of IL-17, IFN-γ protein and mRNA in 27 cases of middle ear cholesteatoma, 23 cases of otitis media granulation and 19 cases of normal external auditory canal skin were detected by immunohistochemistry, ELISA and RT-PCR. And analyze the correlation between its expression and bone destruction of cholesteatoma. Results IL-17 was strongly expressed in the subcutaneous stroma of middle ear cholesteatoma, which was significantly higher than that of the granulation tissue and external auditory canal (P <0.05). IFN- There was a strong positive expression in the stroma, which was significantly higher than that of the external auditory meatus. There was no significant difference in content between the two groups (P> 0.05). The expression of IL-17 mRNA in cholesteatoma was higher than that in granulation tissue and external auditory canal. The expression of IFN-γmRNA in cholesteatoma was higher than that in granulation tissue and external auditory canal (P <0.05) The expression of IL-1 and IFN-γ in ear cholesteatoma was positively correlated with the degree of bone destruction (P <0.01). Conclusion The expression of IL-17 and IFN-γ in cholesteatoma of middle ear is up-regulated, which is related to the degree of bone destruction.
