目的分离并鉴定食管癌细胞株EC/CUHK1的耐砷亚株。 方法胰酶消化法分离亚株EC/CUHK1(As- ) ,2 μmo1/L三氧化二砷 (As2 O3 )诱导凋亡并以流式细胞仪及电子显微镜鉴定。 结果 2 μmo1/LAs2 O3诱导下EC/CUHK1发生明显凋亡 ,亚株EC/CUHK1(As - )无凋亡发生。 结论食管癌细胞株EC/CUHK1中存在耐砷亚株 ,EC/CUHK1和其耐砷亚株EC/CUHK1(As - )可作为研究细胞凋亡敏感性差异机制的适宜模型。
Objective To isolate and identify arsenic-resistant subtypes of esophageal cancer cell line EC/CUHK1. Methods The mycobacterial EC/CUHK1(As-) was isolated by trypsin digestion, apoptosis was induced by 2 μmol/L arsenic trioxide (As2O3) and identified by flow cytometry and electron microscopy. Results The apoptosis of EC/CUHK1 was induced by 2 μmol1/LAs2 O3, and no apoptosis of EC/CUHK1 (As − ) was observed. Conclusion There is an arsenic-tolerant subtype in esophageal cancer cell strain EC/CUHK1. EC/CUHK1 and its arsenic-tolerant subgenomic strain EC/CUHK1(As -) can be used as suitable models to study the mechanism of apoptosis sensitivity.
