目的:研究纤维蛋白原降解产物体外诱导小鼠腹腔巨噬细胞释放白细胞介素-1(IL-1)及白细胞介素-6(IL-6)的作用及一种新型蛋白激酶C(PKC)抑制剂Ro 31-8220(Ro)的影响,方法:胸腺细胞增殖法和B_9细胞增殖MTF法测定IL-1和IL-6活性.结果:纤维蛋白原降解产物促进小鼠腹腔巨噬细胞释放IL-1及IL-6,Ro(0.01-1 μmol·L~(-1))明显抑制IL-I及IL-6的释放,结论:Ro抑制纤维蛋白原降解产物诱导的小鼠腹腔巨噬细胞释放IL-I及IL-6。
Objective: To investigate the effect of fibrinogen degradation products on the release of interleukin-1 (IL-1) and interleukin-6 (IL-6) by mouse peritoneal macrophages and a novel protein kinase C (PKC) Inhibitor Ro 31-8220 (Ro) was determined by MTT assay.Methods: The activity of IL-1 and IL-6 were determined by thymocyte proliferation assay and B9 cell proliferation MTT assay.Results: The fibrinogen degradation products promoted the release of IL from mouse peritoneal macrophages 1 and IL-6, Ro (0.01-1 μmol·L -1) significantly inhibited the release of IL-1 and IL-6. Conclusion: Ro inhibits fibrinogen-induced mouse peritoneal macrophages Release IL-I and IL-6.
