目的 :观察逍遥散对抗慢性应激状态下大鼠海马神经元代细胞内N-甲基-D-天门冬氨酸受体(NR)各亚基m RNA表达的影响。方法:用Taqman荧光定量PCR法测定2-△△Ct相对定量法计算NR各亚基表达量相对值。结果:慢性应激微环境导致海马神经元代细胞内NR的各亚基m RNA表达有不同程度的改变;在拟慢性应激作用前对细胞进行对应血清处理,逍遥散含药血清使NR2Am RNA表达水平明显上调,NR2B m RNA、NR1m RNA表达水平亦上调,作用明显但未达到正常水平。结论:逍遥散治疗血清能改变NR的各亚基m RNA表达,从而达到保护神经元并对抗慢性应激损伤起到一定的作用。
Objective: To observe the effects of Xiaoyaosan on m RNA expression of N-methyl-D-aspartate receptor (NR) subunit in rat hippocampal neurons under chronic stress. Methods: Relative expression of NR subunits was calculated by Taqman fluorescence quantitative PCR method. Results: Chronic stress microenvironment resulted in varying degrees of m RNA expression of NR subunits in hippocampal neurons. Corresponding serums were treated before chronic stress was induced. Xiaoyao powder containing NR2Am RNA The expression level of NR2B m RNA and NR1 m RNA were up-regulated, but the effect was not obvious. Conclusion: Xiaoyaosan treatment serum can change the NR subunit m RNA expression, so as to protect neurons and play a role in the prevention of chronic stress injury.
