目的 探讨体外内毒素 (LPS)对LO2 细胞的直接作用以及N 乙酰半胱氨酸 (N acetylcysteine,NAC)的抑制效果。方法 将LO2 细胞培养 2 4h后换用含LPS(2 0 μg/ml)和 /或NAC(5mmol/L)的新鲜培养液 ,提取肝细胞的基因组DNA ,经琼脂糖凝胶电泳观察肝细胞生物化学性质的改变 ;用透射电镜研究其形态学变化 ;TUNEL标记法检测细胞凋亡。结果 LPS直接作用的LO2 细胞在形态学上表现出胞浆浓缩核凝聚成团块、形成凋亡小体、细胞器结构基本正常等凋亡细胞的特征。将NAC与LPS同时加入培养液 ,作用 16h发现NAC可明显减少因LPS所诱导的LO2 细胞的凋亡。结论 LPS在体外可诱导肝细胞凋亡 ,NAC能抑制其作用。
Objective To investigate the direct effect of endotoxin (LPS) on LO2 cells and the inhibitory effect of N acetylcysteine (NAC). Methods The LO2 cells were cultured for 24 hours and then the cells were extracted with fresh medium containing LPS (20 μg / ml) and / or NAC (5 mmol / L) to extract the genomic DNA of hepatocytes. The hepatocytes were observed by agarose gel electrophoresis Changes in the chemical properties; Transmission electron microscopy morphological changes; TUNEL labeling assay apoptosis. Results LO2 cells directly treated by LPS showed morphologically condensed nuclei of condensed cytoplasm into clumps, forming apoptotic cells with apoptotic bodies and basically normal organelle structure. NAC and LPS were added to the culture medium at the same time, the effect of NAC for 16 h was found to significantly reduce the LPS-induced LO2 cell apoptosis. Conclusion LPS can induce hepatocyte apoptosis in vitro and NAC can inhibit its function.
