目的探讨白色芸豆防御素对肺癌细胞株A549增殖的抑制作用及可能机制。方法构建白色芸豆防御素的真核表达载体pcDNA3.1-defensin-byd,并将其转染至A549细胞中。采用磺酰罗丹明B法检测A549细胞活力,流式细胞术检测细胞凋亡,Western blot检测凋亡蛋白表达。结果白色芸豆防御素可显著抑制A549细胞活性,促进A549细胞凋亡(P<0.05),上调p53及半胱天冬氨酸蛋白酶3(Caspase-3)蛋白表达(P<0.05)。结论白色芸豆防御素通过上调p53及Caspase-3的表达促进A549细胞凋亡,抑制其增殖。
Objective To investigate the inhibitory effect of white kidney bean defensins on the proliferation of lung cancer cell line A549 and its possible mechanism. Methods The eukaryotic expression vector pcDNA3.1-defensin-byd of white kidney bean defensin was constructed and transfected into A549 cells. The viability of A549 cells was detected by sulfonyl rhodamine B method. Apoptosis was detected by flow cytometry and the protein expression of apoptosis was detected by Western blot. Results White kidney bean defensins could significantly inhibit the activity of A549 cells, promote the apoptosis of A549 cells (P <0.05) and upregulate the expression of p53 and Caspase-3 protein (P <0.05). Conclusion White kidney bean defensins promote the apoptosis of A549 cells by up-regulating the expression of p53 and Caspase-3, and inhibiting their proliferation.
