目的:研究丹参酚酸类化合物的体外抗氧化暨对帕金森病细胞模型的神经保护作用。方法:应用MTT比色法检测酚酸类化合物对过氧化氢以及1-甲基-4-苯基吡啶阳离子(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,MPP+)诱导的PC12细胞损伤模型的存活率影响;应用全自动生化分析仪检测活性最强的原儿茶酸(protocatechuic acid,PAc)对MPP+诱导的PC12细胞损伤模型的乳酸脱氢酶释放的影响;应用高内涵筛选系统检测PAc对MPP+诱导的PC12细胞损伤模型的凋亡多参数(F-actin和线粒体膜电位)的影响。结果:Pac 200 mg·L-1的抗氧化活性最强,它可抑制MPP+诱导的乳酸脱氢酶释放和凋亡多参数改变。结论:PAc具有抗PC12细胞氧化损伤作用。
OBJECTIVE: To study the anti-oxidative effects of salvianolic acids in vitro and the neuroprotective effect on Parkinson’s disease cell model. Methods: MTT assay was used to determine the effect of phenolic acids on the induction of hydrogen peroxide and 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPP +) PC12 cell injury model of survival; the application of automatic biochemical analyzer to detect the most active protocatechuic acid (PAc) MPP + -induced PC12 cell damage model of lactate dehydrogenase release; application of high Consensus screening system was used to detect the effect of PAc on the multi-parameters (F-actin and mitochondrial membrane potential) of MPP + -induced injury in PC12 cells. Results: Pac 200 mg · L-1 had the strongest antioxidant activity, which could inhibit MPP + -induced changes of multi-parameters of lactate dehydrogenase release and apoptosis. Conclusion: PAc has anti-oxidative effect on PC12 cells.
