目的比较不同类型宫颈癌细胞株在不同剂量和剂量率射线照射下的放射敏感性特点。方法选取3种宫颈癌细胞株:CaSki、HeLa和SiHa细胞株,建立3种细胞放射敏感性模型,运用克隆形成实验分析不同剂量和剂量率照射下3种细胞的放射敏感性及其差别。结果 3种宫颈癌细胞放射敏感性参数如下:CaSki细胞在2 Gy照射后的存活分数为57.37%,HeLa细胞为70.62%,SiHa细胞为74.77%,三者差异具有统计学意义(P=0.002)。CaSki、HeLa、SiHa细胞在不同剂量率200、400、600 cGy/min照射后的存活分数差异无统计学意义(P>0.05)。结论 3种宫颈癌细胞的放射敏感性以CaSki为最高,HeLa次之,SiHa最低。本研究所用剂量率对3种细胞的存活分数无明显影响。
Objective To compare the radiosensitivity of different types of cervical cancer cell lines under different dose and dose rate radiation. Methods Three cervical cancer cell lines, CaSki, HeLa and SiHa, were selected to establish the radiosensitivity model of three kinds of cells. The radiosensitivity and the difference of the three kinds of cells under different dose and dose rates were analyzed by colony formation assay. Results The radiosensitivity parameters of three kinds of cervical cancer cells were as follows: the survival rate of CaSki cells was 57.37% after 2 Gy irradiation, 70.62% for HeLa cells and 74.77% for SiHa cells, the difference was statistically significant (P = 0.002) . The survival scores of CaSki, HeLa and SiHa cells after 200, 400 and 600 cGy / min irradiation were not significantly different (P> 0.05). Conclusion The radiosensitivity of the three kinds of cervical cancer cells is highest with CaSki, followed by HeLa and lowest with SiHa. The dose rate used in this study had no significant effect on the survival of the three cell types.
