目的建立黄药子散中黄芩苷含量测定的HPLC方法。方法色谱柱为NUCLEODUR C18Pyramid(250 mm×4.6mm,5μm),流动相为乙腈-水(26∶74,含0.5%三乙胺,磷酸调p H 3.0),流速为1 ml·min-1,检测波长为278 nm,进样量为10μl,柱温为30℃。结果回归方程为Y=31.70 X-23.00,r=0.9999,黄芩苷的浓度线性范围是7.72~193.0μg·ml-1。平均加样回收率为101.4%,RSD为1.4%(n=6)。结论该方法操作简便、稳定性好,可用于黄药子散中黄芩苷的含量测定。
Objective To establish HPLC method for the determination of baicalin in Radix Astragali. Methods The chromatographic column was NUCLEODUR C18Pyramid (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-water (26:74, containing 0.5% triethylamine and phosphoric acid) and the flow rate was 1 ml · min- The detection wavelength was 278 nm, the injection volume was 10μl and the column temperature was 30 ℃. Results The regression equation was Y = 31.70 X-23.00, r = 0.9999. The linear range of baicalin was 7.72-193.0 μg · ml-1. The average recovery was 101.4% with a RSD of 1.4% (n = 6). Conclusion The method is simple, stable, and can be used for the determination of baicalin in Huang Yao Zi San.
