目的 探讨阿尔茨海默病 (AD)、帕金森病 (PD)、帕金森病痴呆 (PDD)的发病机制及与载脂蛋白E (ApoE)基因多态性的关系。方法 对AD组 (48例 )、PD组 (5 4例 )、PDD组 (43例 )和非痴呆对照组(2 34例 )的ApoE基因频率及基因型分布进行对照研究。结果 AD组ApoEε4基因频率明显升高 (2 1 9% ) ,显著高于非痴呆对照组 (5 1% ,P <0 0 1) ,表明ε4与AD发病关系极为密切。PD和PDD组ApoEε4和ε2基因频率与非痴呆对照组相比无显著差异 (P >0 0 5 ) ,提示ε4和ε2与PD、PDD发病无关。结论 ApoEε4是导致AD发病的易感或危险基因之一 ,而与PD和PDD的发病无关 ;AD和PD、PDD的发病机理不同。
Objective To investigate the pathogenesis of Alzheimer’s disease (AD), Parkinson’s disease (PD) and Parkinson’s disease (PDD) and its relationship with apolipoprotein E (ApoE) gene polymorphism. Methods ApoE gene frequency and genotype distribution in AD group (48 cases), PD group (54 cases), PDD group (43 cases) and non-demented control group (234 cases) were compared. Results The frequency of ApoEε4 gene was significantly higher in AD group (21.9%) than that in non-demented control group (51%, P <0.01), indicating that ε4 is closely related to AD. The frequencies of ApoEε4 and ε2 genes in PD and PDD group were not significantly different from those in control group (P> 0.05), which suggested that ε4 and ε2 were not associated with PD and PDD. Conclusion ApoEε4 is one of the susceptible or dangerous genes that cause AD, but not to the pathogenesis of PD and PDD. The pathogenesis of AD, PD and PDD are different.
