目的观察骨癌痛模型大鼠患侧腰段脊髓STAT3的表达,探讨其在骨癌痛中的作用机制。方法选择雌性SD大鼠20只,随机分为2组(n=10):模型组,为左侧胫骨上段骨髓腔注入3μLMADB-106大鼠乳腺癌细胞(4.8×103/μL);对照组,为左侧胫骨上段骨髓腔注入3μLHank液。第14天,采用免疫组织化学方法和荧光双标法观察两组大鼠患侧腰段脊髓STAT3和GFAP免疫反应阳性产物的变化和关系。结果对照组大鼠对机械痛刺激的缩爪阈值在术后各时间点组内相比差异无显著性;模型组大鼠在术后缩爪阈值与对照组相比差异有显著性(P<0.05)。左侧胫骨上段骨髓腔注入癌细胞后第14d,骨癌痛模型大鼠患侧脊髓STAT3、GFAP免疫反应阳性产物表达明显增加(P<0.05);荧光双标免疫组化显示患侧脊髓STAT3与星形胶质细胞标志物GFAP有共表达。结论脊髓背角STAT3参与了骨癌痛的发生和发展,脊髓星形胶质细胞在骨癌痛中可能通过JAK/STAT3信号通路发挥作用。
Objective To observe the expression of STAT3 in the lumbar spinal cord of the rat model of bone cancer pain and to explore its mechanism of action in bone cancer pain. Methods Twenty female Sprague-Dawley rats were randomly divided into 2 groups (n = 10): model group, injected 3μL of MADB-106 rat breast cancer cells (4.8 × 103 / μL) into the left medial tibial bone marrow; Into the left tibia medullary cavity marrow cavity 3μL Hank solution. On the 14th day, the changes and relationships of STAT3 and GFAP immunoreactive products in the affected side lumbar spinal cord of the two groups were observed by immunohistochemistry and fluorescent double labeling. Results There was no significant difference between the control group and the control group in the paw withdrawal threshold of the mechanical pain stimulation in the postoperative time points (P < 0.05). On the 14th day after injection into the left medial tibial bone marrow cavity, the expression of STAT3 and GFAP immunoreactive products in the ipsilateral spinal cord increased significantly in the bone cancer pain model rats (P <0.05). Double immunofluorescence staining showed that STAT3 and Astrocyte marker GFAP is co-expressed. Conclusions STAT3 is involved in the occurrence and development of bone cancer pain in spinal dorsal horn. Spinal cord astrocytes may play a role in the pain of bone cancer through JAK / STAT3 signaling pathway.
