目的研究浸润性子宫颈癌(infiltrative cervical cancer,ICC)及子宫颈上皮内瘤变(intraepithelial neoplasia,CIN)中人乳头瘤病毒(human papilom avirus,HPV)感染率及型别分布。方法选取重庆市肿瘤医院组织病理学诊断为CINⅡ、CINⅢ级和宫颈鳞癌的组织标本58例作为研究对象,SPF10LiPA25及HPV DNA酶免疫检测法(DNA enzyme immunoassay,DEIA)进行DNA的提取及扩增;采用线形探针反向杂交技术检测病变组织中HPV型别。结果 58例标本中有56例检测出HPV感染,HPV感染率96.55%,其中CINⅡ、CINⅢ级和子宫颈癌中HPV感染的阳性率分别为100.00%、100.00%、95.00%。共检测出13种HPV型别,HPV16感染率最高(60.00%),其次为HPV18(7.69%)、HPV58(7.69%)和HPV31(4.62%)。宫颈鳞癌、CINⅢ、CINⅡ级HPV多重感染率分别为6.06%、26.31%、33.33%。41~50岁高危型HPV感染率最高(44.64%)。结论 HPV16为重庆市最常见的HPV感染型别,HPV18、HPV58和HPV31也有较高感染率。
Objective To study the infection rate and type distribution of human papilom avirus (HPV) in infiltrative cervical cancer (ICC) and cervical intraepithelial neoplasia (CIN). Methods Tissue specimens of CINⅡ, CINⅢ and squamous cell carcinoma of the cervix in Chongqing Tumor Hospital were selected as the research objects. SPF10LiPA25 and HPV DNA immunoassay (DEIA) were used to extract and amplify DNA The HPV type in diseased tissues was detected by linear probe reverse hybridization. Results The HPV infection rate was 96.55% in 56 cases of 58 samples. The positive rates of HPV infection in CINⅡ, CINⅢ and cervical cancer were 100.00%, 100.00% and 95.00% respectively. Thirteen HPV types were detected with the highest infection rate of HPV16 (60.00%), followed by HPV18 (7.69%), HPV58 (7.69%) and HPV31 (4.62%). Cervical squamous cell carcinoma, CIN Ⅲ, CIN Ⅱ HPV multiple infection rates were 6.06%, 26.31%, 33.33%. The highest infection rate of high-risk HPV in 41-50 years old (44.64%). Conclusion HPV16 is the most common type of HPV infection in Chongqing. HPV18, HPV58 and HPV31 also have a higher infection rate.
