目的:采用病、证模型复合的方法建立血瘀型颈椎病病证结合动物模型。方法:选择3月龄雌性SPF级SD大鼠30只,随机分为正常对照组、颈椎病模型组和血瘀型颈椎病模型组,每组10只。颈椎病模型采用动静力失衡性大鼠颈椎间盘退变模型,血瘀型颈椎病模型在动静力失衡性大鼠颈椎间盘退变模型基础上复合肾上腺皮质激素加肾上腺素法血瘀模型制成。通过动物体征、血液流变学、CD62p验证血瘀证;通过颈椎间盘组织病理学、Ⅱ型和X型胶原的免疫组织化学检测及Agc1、Col2a1、MMP-13和TIMP-1的基因表达评判椎间盘退变程度证实颈椎病模型的成立。结果:血瘀型颈椎病模型组动物舌质瘀紫、瘀斑,尾色瘀青;血液流变学指标及CD62p均有不同程度增高,颈椎间盘组织病理学观察可见退变更加明显,Ⅱ型胶原蛋白表达减少,X型胶原表达增高,与正常组和颈椎病组相比差异均有统计学意义(P<0.05或P<0.01);Agc1、Col2a1和TIMP-1的基因表达降低,MMP-13基因表达增高,Agc1、Col2a1的差异与正常组比较有统计学意义(P<0.05或P<0.01),与颈椎病组比较无统计学意义。结论:通过病、证模型复合的方式建立了大鼠血瘀型颈椎病模型,血瘀可以加重颈椎间盘的退变。
OBJECTIVE: To establish a combined animal model of cervical syndrome with blood-stasis syndrome by syndrome differentiation and syndrome differentiation. Methods: Thirty female Sprague-Dawley rats of 3 months old were randomly divided into normal control group, cervical spondylosis model group and cervical spondylosis model group with 10 rats in each group. Cervical spondylosis model with dynamic and static imbalance of cervical disc degeneration model, blood stasis type cervical spondylosis model in the dynamic and static imbalance of cervical disc degeneration model based on adrenal cortical hormone and adrenaline blood stasis model made. Blood stasis syndrome was confirmed by animal signs, hemorheology and CD62p. The expression of Agc1, Col2a1, MMP-13 and TIMP-1 was evaluated by cervical histopathology, immunohistochemistry of type Ⅱ and type X collagen, Degeneration confirmed the establishment of cervical spondylosis model. Results: The blood stasis type cervical spondylosis model group tongue tongue bruise, ecchymosis, tail color bruise; hemorheology and CD62p all increased to varying degrees, cervical disc tissue pathology showed more obvious degeneration, type Ⅱ (P <0.05 or P <0.01). The expressions of Agc1, Col2a1 and TIMP-1 were down-regulated while the expressions of MMP- 13 gene expression increased, Agc1, Col2a1 differences compared with the normal group was statistically significant (P <0.05 or P <0.01), and cervical spondylosis group was not statistically significant. Conclusion: The model of cervical spondylosis of blood stasis in rats was established by means of combination of disease and syndrome model. Blood stasis can aggravate the degeneration of cervical intervertebral disc.
