目的:探讨罗格列酮(RSG)对高糖环境下人肾小管上皮细胞(HK-2)血小板反应蛋白1(TSP-1)和骨形成蛋白7(BMP-7)mRNA和蛋白表达的影响。方法:将体外培养的人肾小管上皮细胞分为正常糖对照组(NG)、高糖组(HG)、渗透浓度对照组(MG)、分别含RSG5、10、20μmol/L的高糖DMEM培养基培养的RSG干预组(R5、R10、R20),并于培养48h后终止培养。免疫细胞化学方法检测人肾小管上皮细胞TSP-1、BMP-7、结缔组织生长因子(CTGF)蛋白的表达,RT-PCR法检测人肾小管上皮细胞中TSP-1、BMP-7、CTGFmRNA表达。结果:高糖环境下,HK-2细胞TSP-1、CTGFmRNA和蛋白表达明显升高(P<0.05),BMP-7mRNA和蛋白表达明显降低(P<0.05),加入RSG后,HK-2细胞TSP-1、CTGF的表达较NG组明显降低(P<0.05),BMP-7的表达明显升高(P<0.05)。结论:RSG可以抑制高糖诱导的人肾小管上皮细胞TSP-1、CTGF高表达,提高BMP-7的表达,具有一定的肾脏保护作用。
AIM: To investigate the effect of rosiglitazone (RSG) on mRNA and protein expression of TSP-1 and BMP-7 in human renal tubular epithelial cells (HK-2) under high glucose conditions . Methods: Human renal tubular epithelial cells cultured in vitro were divided into normal glucose control group (NG), high glucose group (HG) and osmotic concentration control group (MG), and cultured in high glucose DMEM containing RSG 5,10 and 20μmol / L RSG intervention groups (R5, R10, R20) were cultured in vitro and cultured after 48 hours. The expressions of TSP-1, BMP-7 and CTGF in human renal tubular epithelial cells were detected by immunocytochemistry. The expressions of TSP-1, BMP-7 and CTGF mRNA in human renal tubular epithelial cells were detected by RT-PCR . Results: The mRNA and protein expressions of TSP-1 and CTGF in HK-2 cells were significantly increased (P <0.05), and the mRNA and protein expressions of BMP-7 in HK-2 cells were significantly decreased The expression of TSP-1 and CTGF was significantly lower than that of NG group (P <0.05), and the expression of BMP-7 was significantly increased (P <0.05). CONCLUSION: RSG can inhibit high glucose-induced high expression of TSP-1 and CTGF in human renal tubular epithelial cells and improve the expression of BMP-7, which has a certain renal protective effect.
